The Ancient Cave from Lufia II, if you want to count it...again. On a slightly more serious note, I guess Darm Tower from Ys I could count by virtue of being basically half the entire game.

Many of the games in the mystery dungeon series have 100 floor dungeons in them at the end of the games. Zettai Hero Project is another with ludicrously long dungeons.

There is also the 100 floor dungeon in tactics ogre let us cling together that you have to do 3 or 4 times to get everything. it takes a couple days of play sessions to get through it once!

There's a bunch of RPGs with 100-floor bonus dungeons, but the longest main-game dungeon I've seen is the final in FF4: The After Years. It's completely absurd.

The "Bladder of Steel" Achievement in Rock Band 2 requires you to play the entire setlist of the game in a row, without failing any songs...and without pausing. It takes over six hours to complete, and you basically need a bucket in front of you the whole time.

In the original Japanese release of Persona there was a quest called the Snow Queen Quest. I think it takes about ten hours to complete and there is only one save point. Not sure if it was included in the US PSP rerelease. I know it was mentioned before but Hell’s Gate in Tactics Ogre is brutal. I think Atlus sent out t-shirts to people who completed it twice.

Truly someone who hates their player base. If you think that's bad, 7 Day Survivor in Dead Rising requires you to play for ~14 hours consecutively with no saves, as well.

Maybe someone who could already tell that gamers take challenges very differently if they're tied to an achievement vs. if they aren't? Most players wouldn't even want to attempt it, so the 'player base' in question becomes a) people who are into that sort of thing (in which case there's no hate to be had) and b) people who are weird about achievements (in which case: got 'em!) Stuff like that is pretty funny to me.

Stuff like that is pretty funny to me. Yeah. I did 7 Day Survivor (even though it destroyed my X360) and lived to tell the tale. That's a real achievement, not something where you just have to grind or search for collectibles with a FAQ or do the bare minimum to complete the game. If I designed achievements/trophys/whatever, they'd be like that. Sadistic, insane stuff to make people go insane but be proud of it.

I never tried it myself because the mode really did not seem fun.

The longest final dungeon I've ever gone through in an RPG has to be Kagutsuchi from Shin Megami Tensei: Nocturne. I spent a good 15 hours in there. It was seriously like a game all its own.
As far as non-RPGs go, in recent memory, the indie game "The Sword of IANNA" from just last year had pretty insanely huge levels. It's a Prince of Persia-style cinematic platformer, and you only get passwords between levels (of which there are 8, with a secret 9th level unlockable as well), but you have to be ready and willing to really sit down and put in some serious playtime each time you enter a password, because your first run through any of those stages will likely last you at least one full hour. And bear in mind, this is a homebrew game for 8-bit microcomputers, so the average stage length among its brethren is 5-10 minutes, tops -- and hell, even 10 minutes would feel kind of long! So these hour-long stages in Sword of IANNA make it a really daunting, but also really rewarding, game to play.
